目的 探讨年龄对脑缺血的影响,建立一个稳定的、与人类卒中年龄相关的老年大鼠局灶性脑缺血模型。方法 采用Watson法制作光化学脑缺血模型,对比观察老年鼠及青年鼠缺血诱导后血脑屏障、梗塞灶大小及光镜、电镜下缺血区的病理变化。结果 (1)缺血诱导的大鼠存活率100%,梗塞灶恒定出现。(2)与青年鼠相比,缺血老年鼠的精神差、血脑屏障破坏重、梗塞灶大,神经细胞受损、炎性细胞浸润严重而胶质细胞及微血管增生等组织修复反应轻。结论 同等缺血打击下,老年鼠的缺血性损害重、修复能力低。光化学诱导老年大鼠局灶性脑梗塞模型稳定性好、死亡率低,是研究脑缺血相对理想的模型。“,”Objective To study the effects of age on cerebral ischemi a,we developed a stable model of focal cerebral ischemia in aged rats which is a ge-related with human stroke.Methods Cerebral ischemia was induced photochemically in young and aged rats according to the procedure described by Waston.The observati ons of blood-brain barrier(BBB),infarct size and pathological changes under lig h t and transmission electron microscopes were compared between the two groups aft er ischemic induction.Results (1) The survival rate of ischemic rates was 100% a nd the infarct lessions were reproducible.(2) The state of mind,BBB and neurol ogical cells damage and infiltration of inflammatory cells were worse,and the i nfarct size was large,while the response of tissue repair was slighter in aged rats compared to young rats.Conclusions The neurological impairment is more significa nt and repair capability lower in aged rats with respect to young rats.The phot ochemical infarct model in aged rats is a relatively perfect model to investigat e cerebral ischemia.
