目的探讨肿瘤坏死因子受体相关因子-5(tumor necrosis factor receptor-associated factor-5,TRAF-5)在炎症性肠病患者外周血单个核细胞和血浆的表达,比较炎症性肠病患者TRAF-5表达和正常对照者的差异,分析血浆TRAF-5表达与内镜下疾病活动性的相关性。方法应用酶联免疫吸附试验(enzyme-linked immuno sorbent assay,ELISA)分析炎症性肠病患者和正常对照者血浆中TRAF-5蛋白的表达,SYBR-green real time PCR方法分析炎症性肠病患者和正常对照者外周单个核细胞中TRAF-5 mRNA的表达。数据处理使用GraphPad Prism 5。结果克罗恩病患者(P=0.003)和溃疡性结肠炎患者(P=0.000)血浆TRAF-5水平显著高于正常对照者,但是克罗恩病患者(r=-0.167,P=0.194)以及溃疡性结肠炎患者(r=0.008,P=0.952)血浆TRAF-5表达水平和内镜下疾病活动指数均无显著的相关性。克罗恩病患者(P=0.000)和溃疡性结肠炎(P=0.000)患者外周血单个核细胞TRAF-5 mRNA表达均显著高于正常对照者。结论炎症性肠病患者血浆和外周血单个核细胞中均存在TRAF-5的激活,但血浆TRAF-5的高表达不能反映内镜下疾病的活动程度。
Objective To investigate the expression of tumor necrosis factor receptor-associated factor-5 (TRAF-5) in peripheral blood mononuclear cells and plasma in patients with inflammatory bowel disease. To compare the expression of TRAF- 5 expression and normal controls, analyze the correlation between plasma TRAF-5 expression and endoscopic disease activity. Methods The expression of TRAF-5 protein in plasma of patients with inflammatory bowel disease and normal controls was analyzed by enzyme-linked immunosorbent assay (ELISA). SYBR-green real time PCR method was used to analyze the expression of TRAF- Expression of TRAF-5 mRNA in peripheral blood mononuclear cells of normal controls. Data processing uses GraphPad Prism 5. Results Plasma levels of TRAF-5 in Crohn’s disease patients (P = 0.003) and ulcerative colitis patients (P = 0.000) were significantly higher than those in normal controls, but Crohn’s disease patients (r = -0.167, P = 0.194) (R = 0.008, P = 0.952). There was no significant correlation between the level of TRAF-5 expression in plasma and the activity index of endoscopic disease in patients with ulcerative colitis. The expression of TRAF-5 mRNA in peripheral blood mononuclear cells in patients with Crohn’s disease (P = 0.000) and ulcerative colitis (P = 0.000) were significantly higher than those in normal controls. Conclusion The activation of TRAF-5 exists in both plasma and peripheral blood mononuclear cells in patients with inflammatory bowel disease. However, the high expression of TRAF-5 in plasma does not reflect the degree of endoscopic activity.
