目的:探讨大黄素对人结肠癌细胞株RKO、Caco-2的体外增殖的抑制作用。方法:MTT比色法检测细胞活力;倒置显微镜显像观察细胞数量变化;FCM检测细胞凋亡率及周期分布;Caspase-3分光光度法检测Caspase-3酶蛋白活性。结果:随着作用时间的延长以及大黄素浓度的升高,大黄素对人结肠癌RKO、Caco-2细胞的体外增殖抑制作用增高;大黄素阻滞两种细胞于S期,使G2/M期细胞比率明显下降,激活Caspase-3酶蛋白;提高细胞凋亡率。结论:大黄素抑制人结肠癌细胞株RKO、Caco-2体外增殖,其作用呈浓度、时间依赖性;大黄素能够阻滞RKO、Caco-2细胞株细胞周期并激活Caspase-3诱导细胞凋亡。
Objective: To investigate the inhibitory effect of emodin on human colon cancer cell lines RKO and Caco-2 in vitro. Methods: Cell viability was detected by MTT colorimetric assay. The number of cells was observed by inverted microscope. The apoptosis rate and cell cycle distribution were detected by FCM. Caspase-3 activity was detected by Caspase-3 spectrophotometry. RESULTS: With the prolongation of action time and the increase of emodin concentration, emodin inhibited the proliferation of human colon cancer RKO and Caco-2 cells in vitro. Emodin blocked the proliferation of both cells in S phase and G2 / M The percentage of stage cells decreased significantly, which activated Caspase-3 enzyme protein and increased the rate of apoptosis. CONCLUSION: Emodin can inhibit the proliferation of human colon cancer cell lines RKO and Caco-2 in a concentration-dependent and time-dependent manner. Emodin can block the cell cycle of RKO and Caco-2 cell lines and induce apoptosis of Caspase-3 cells .
