目的研究温州市瓯海地区宿主携带汉坦病毒的状况和型别。方法经间接免疫荧光(IFA)试验初筛为阳性的鼠肺标本,用巢式RT-PCR扩增阳性标本中汉坦病毒的S基因片段上的目的核苷酸序列(620~999nt),并以ClustalX(1.83)和Phylip3.63构建系统进化树进行分型和分子进化分析。结果温州市瓯海地区共捕获啮齿动物148只,5份鼠肺标本中汉坦病毒抗原阳性,其中褐家鼠3只,黄胸鼠2只,病毒携带率为3.38%,与汉坦病毒分型标准比较,均属于SEOV型。结论瓯海地区褐家鼠和黄胸鼠携带S1和S3亚型汉坦病毒,显示出汉坦病毒的多样性。
Objective To study the status and type of Hantaan virus carried by Ouhai area in Wenzhou City. Methods The positive rat lung specimens were detected by indirect immunofluorescence (IFA). The target nucleotide sequence (620-999 nt) of the S gene fragment of Hantavirus in positive specimens was amplified by nested RT-PCR. The phylogenetic tree was constructed using ClustalX (1.83) and Phylip3.63 for typing and molecular evolution analysis. Results A total of 148 rodents were caught in the Ouhai area of Wenzhou City. Among the 5 samples, 3 were Rattus norvegicus, 2 Rattus flavipectus and 2 Rattus flavipectus. The virus carrying rate was 3.38% Type standard comparison, are SEOV type. Conclusion Rattus norvegicus and Rattus flavipectus carry the Hantavirus strains S1 and S3 in Ouhai area, indicating the diversity of Hantavirus.
