目的分离培养大鼠骨髓间充质干细胞(bone marrow stromal stem cells,BMSCs)并进行慢病毒载体介导的基因转染研究。方法利用密度梯度离心法分离、纯化、扩增培养大鼠骨髓间充质干细胞。以脂质体法进行慢病毒感染大鼠BMSCs。结果分离培养出大鼠BMSCs,并用携带绿色荧光蛋白(GFP)报告基因的慢病毒成功感染大鼠BMSCs。结论获得大鼠BMSCs并进行慢病毒载体介导的基因修饰,为骨髓间充质干细胞基因治疗研究奠定了基础。
Objective To isolate and culture rat bone marrow stromal stem cells (BMSCs) and study the lentiviral vector-mediated gene transfection. Methods The rat bone marrow mesenchymal stem cells were isolated and purified by density gradient centrifugation. Infection of BMSCs with lentivirus in rat liposomes. Results Rat BMSCs were isolated and cultured, and BMSCs were successfully infected with lentivirus carrying green fluorescent protein (GFP) reporter gene. Conclusion Obtaining rat BMSCs and carrying out lentiviral vector-mediated gene modification laid the foundation for gene therapy of bone marrow mesenchymal stem cells.
