对单纯用二乙基亚硝胺 (DENA)及氯化钆 (GC)或酵母多糖 (ZM)分别阻断或激活 Kupffer细胞后 ,同时给以 DENA所引起的大鼠肝癌中的增殖细胞核抗原 (PCNA )、 bax、 p5 3蛋白的表达和肝癌细胞的凋亡运用免疫组织化学和 TU NEL 法进行了对比研究 ,以探讨 Kupffer细胞在大鼠实验性肝癌细胞凋亡中的作用。结果显示 :肝癌组织的增殖指数、凋亡指数在 C组、 A组、 B组分别呈增高或降低趋势 ;bax、 p5 3阳性率在 C组明显高于 A组 (P<0 .0 5 ) ,同时 ,bax、p5 3蛋白的表达与凋亡指数存在正相关。结果提示 ,Kupffer细胞可能促进实验性肝癌细胞凋亡。
After blocking or activating Kupffer cells with diethylnitrosamine (DENA) and cesium chloride (GC) or zymosan (ZM), respectively, the proliferating cell nuclear antigen in rat liver cancer caused by DENA is simultaneously given ( The expression of PCNA, bax, and p53 protein and the apoptosis of hepatoma cells were compared by immunohistochemistry and TU NEL assays to investigate the role of Kupffer cells in the apoptosis of rat hepatoma cells. The results showed that the proliferation index and apoptotic index of hepatocellular carcinoma were increased or decreased in group C, group A and group B respectively; the positive rates of bax and p53 were significantly higher in group C than in group A (P<0.05). At the same time, there was a positive correlation between the expression of bax and p53 protein and apoptotic index. The results suggest that Kupffer cells may promote apoptosis of experimental hepatoma cells.
