龙葵碱对小鼠睾丸支持细胞波形蛋白表达的影响

目的探讨龙葵碱对小鼠睾丸支持细胞波形蛋白表达的影响。方法将40只雄性昆明小鼠随机分为4组,A组腹腔注射等剂量0.9%NaCl,B、C、D组分别腹腔注射龙葵碱5、10和20 mg/kg,连续染毒14 d,取出睾丸组织。利用苏木素-伊红(HE)染色、睾丸重量、脏器系数的改变观察小鼠睾丸的病理学变化;利用免疫组织化学法检测睾丸波形蛋白的表达和变化。结果 实验表明,与对照组相比,20 mg/kg组的小鼠睾丸组织切片病理变化明显,曲细精管明显萎缩,大量细胞脱落至管腔及管腔外面,生精细胞与支持细胞分离,管腔内细胞排列紊乱,精子生成数大量减少;10和20mg/kg组的小鼠睾丸脏器系数(0.346±0.020、0.222±0.050)明显低于对照组(0.416±0.065),结果具有统计学意义(P<0.05);20 mg/kg组小鼠睾丸波形蛋白表达明显下降,排列无序,阳性支持细胞脱落至曲细精管官腔之中。结论在本试验条件下,龙葵碱可能通过影响支持细胞波形蛋白的表达产生雄性生殖毒性。 Objective To investigate the effect of solanine on the expression of vimentin in mouse testis supportive cells. Results The experimental results showed that compared with the control group, the pathological changes of testes in 20 mg / kg group were obvious, the seminiferous tubules obviously atrophied, a large number of cells were shed to the outside of the lumen and lumen, and the spermatogenic cells were separated from the supporting cells , The intraluminal cells arranged in disorder, a significant reduction in the number of sperm production; 10 and 20mg / kg group of mice testis organ coefficient (0.346 ± 0.020,0.222 ± 0.050) was significantly lower than the control group (0.416 ± 0.065), the results with statistics (P <0.05). In the 20 mg / kg group, the expression of vimentin in the testes decreased significantly and the disordered arrangement of the positive cells dropped into the lumen of the seminiferous tubules. Conclusion Solanine may produce male reproductive toxicity by affecting the expression of vimentin in this assay.
