实验性雷公藤中毒的超微病理学研究

中草药雷公藤又名黄藤、菜虫药、断肠草,是广泛生长在我国长江流域以南各省的一种有毒植物。近年来发现此药除在治疗风湿性关节炎方面有显著疗效外,还在抗炎、抑制体液免疫、治疗白血病及抗肿瘤方面有一定作用。国内已见有关药物成分分析及临床疗效观察的报导,但对病理学方面报告甚少,在超微病理方面尚未见报导。本文用雷公藤根皮对大白鼠进行急性中毒实验,设想从超微结构水平探讨中毒机理,为正确用药提供依据。[材料和方法]实验用Wistar大白鼠40只,随机分成盐水对照组和3个不同剂量实验组,进行急性中毒实验。按中毒剂量陆续处死动物,取心、肝、肾和脾组织常规处理制成超薄切片,电镜观察。[结果和讨论]结果发现各组织均显示不同程度的病理变化,以心组织和脾组织最为突出,肾组织也较为明显,肝组织相对较轻。心肌主要表现 The Chinese herbal medicine Tripterygium wilfordii, also known as yellow rattan, vegetable insecticide and heartbroken grass, is a poisonous plant widely grown in the provinces south of the Yangtze River basin in China. In recent years, the drug has been found to have a significant effect in the treatment of rheumatoid arthritis, but also in the anti-inflammatory, humoral immunity, treatment of leukemia and anti-tumor. Domestic reports on the analysis of pharmaceutical ingredients and observations of clinical efficacy have been reported. However, few reports on pathology have been reported. No report has been made on ultrastructural pathology. In this paper, acute poisoning experiments were conducted on rats with root cane of Tripterygium wilfordii, and the mechanism of poisoning was explored from the ultrastructural level to provide the basis for correct drug use. [Materials and Methods] Forty Wistar rats were randomly divided into saline control group and three different experimental groups for acute poisoning experiments. The poisoned doses were successively sacrificed, and the cortical, liver, kidney and spleen tissues were routinely processed into ultrathin sections and observed under electron microscope. [Results and discussion] The results showed that all tissues showed different degrees of pathological changes, with the most prominent cardiac and splenic tissues, more pronounced renal tissue, and relatively lighter liver tissue.
