本文采用RAPD技术对节节麦、光稃野燕麦及其杂交后代进行分析,目的是为了对它们进行分子标记,确定杂种的真实性。本次实验共选用72个随机引物对节节麦、光稃野燕麦及其杂种进行一次性“扫描”,发现有37个引物能够扩增出节节麦和光稃野燕麦特异的多态性DNA片段,其中包括只在节节麦和杂种中出现的特异DNA片断,只在光稃野燕麦和杂种中出现的特异DNA片断,以及同时出现在节节麦和光稃野燕麦的特异性DNA片段。第一种情况约占29%,第二种情况约占17%,第三种情况约占5%。
In this paper, RAPD analysis of Artemisia scoparia, Oryza rufipogon and their hybrid progeny were analyzed in order to molecular markers of them, to determine the authenticity of hybrids. In this experiment, a total of 72 random primers were used to conduct a one-time “scan” of O. niloticum, O. japonica and their hybrids, and 37 primers were found to amplify specific polymorphic DNA Fragments, including specific DNA segments that occur only in Aegilops tauschii and hybrids, specific DNA segments that occur only in the oyster oats and hybrids, and specific DNA segments that occur simultaneously in the oat and wild oats. The first case accounted for about 29%, the second case about 17%, the third case about 5%.
