目的:优选聚酰胺纯化薄荷总黄酮的工艺条件。方法:以总黄酮含量和吸附-洗脱率为指标,采用单因素试验考察5种不同类型的纯化材料对总黄酮的吸附能力,确定纯化材料,并对其纯化工艺中吸附条件、除杂条件和洗脱条件进行优化。结果:优化工艺条件为上样液质量浓度0.1 g.mL-1,上样量为生药量与聚酰胺体积比1∶6,吸附流速2 BV.h-1,柱床径高比1∶9,待吸附结束后,用3 BV 10%乙醇洗脱除杂,除杂流速4 BV.h-1,再用3 BV 90%乙醇以8 BV.h-1流速洗脱,经聚酰胺(30~60目)纯化后,薄荷总黄酮纯度可达50%。结论:优选的纯化工艺分离效果良好、操作简单、重复性好,可用于分离富集薄荷总黄酮。
OBJECTIVE: To optimize the conditions for the purification of total flavonoids from peppermint by polyamide. Conclusion: The optimized purification process has the advantages of good separation effect, simple operation and good repeatability, and can be used for separation and enrichment of mint flavonoids.
