研究细胞内组胺在PAF诱导血小板活化中的作用.方法:采用血小板聚集及放免法测定TXB2,Fura2负载测细胞内钙.结果:美吡拉敏高于100μmol·L-1抑制PAF诱导的聚集,而西咪替丁在400μmol·L-1亦无效,组胺及2thiazolylethylamine也无协同作用.但在皂素(8-10mg·L-1)通透的血小板,组胺可减弱美吡拉敏的抑制效应,美吡拉敏还可抑制PAF引起的TXB2产生和细胞内钙增加.结论:PAF诱导的血小板活化伴随有细胞内组胺的合成或释放通过TXB2产生和细胞内钙增加这一共同通路.
To study the role of intracellular histamine in the PAF-induced platelet activation. Methods: Platelet aggregation and radioimmunoassay TXB2, Fura 2 load measured intracellular calcium. Results: Pymetrozine above 100 μmol·L-1 inhibited PAF-induced aggregation, whereas cimetidine was also ineffective at 400 μmol·L-1. Histamine and 2thiazolylethylamine also showed no synergistic effect. However, platelets with saponin (8-10 mg · L-1) permeabilized showed that histamine attenuated the inhibitory effect of mepyramine, while mepyramine also inhibited PAF-induced TXB2 production and intracellular calcium increase. CONCLUSION: PAF-induced platelet activation is accompanied by intracellular histamine synthesis or release through the common pathway of TXB2 production and increased intracellular calcium.
