用1%DOC(脱氧胆酸钠)溶解的膜蛋白,经Sephgarose4B柱层析,在外水体积出现合有胰岛素受体活性的蛋白峰。这个单一的蛋白峰经6M盐酸胍处理后,重过用1M盐酸胍平衡的Sepharose 4B柱,可分出四个280毫微米的吸收峰,除了在外水体积的第一个蛋白峰有结合胰岛素活性外,第四个吸收峰也有明显的结合活力。用凝胶电泳也证实了这个小分子活性单位的存在.经分子筛测定,小分子的活性单位分子量约5万道尔顿.
Membrane proteins dissolved in 1% DOC (sodium deoxycholate) were subjected to Sephgarose 4B column chromatography to reveal protein peaks with insulin receptor activity in the external water volume. This single protein peak was resolved on a Sepharose 4B column equilibrated with 1 M guanidine-HCl after treatment with 6 M guanidine-HCl, yielding four 280 nm absorbance peaks except for the first protein peak in the external water volume with bound insulin activity In addition, the fourth absorption peak also has obvious combination of vitality. Gel electrophoresis also confirmed the existence of this small molecule activity unit.According to molecular sieve determination, small molecule activity unit molecular weight of about 50,000 daltons.
