目的:观察祛风宣肺汤对脂多糖(lipopolysaccharide,LPS)引起的感染后咳嗽(postin-fectious cough,PIC)小鼠动物模型咳嗽反射敏感性性和神经源性炎症的治疗作用,并探索可能的机制。方法:雄性的ICR小鼠24只,按数字随机法随机分成空白组、LPS组和中药组。每组8只,通过腹腔注射一定浓度LPS造成小鼠感染后咳嗽的气道高反应性模型,其中中药组并给予中药组祛风宣肺汤灌胃。氨水刺激后人工数小鼠咳嗽次数,肺功能仪测定气道反应性,免疫组化检测小鼠肺中瞬时感受器电位香草酸受体1(transient receptor potential vanilloid 1,TRPV1)表达位置和半定量,蛋白质印迹法(Western Blot,WB)方法检测肺组织中辣椒素受体TRPV1蛋白表达,实时定量PCR(real-time PCR)方法检测肺组织中TRPV1、P物质受体神经肽受体1(NKl)的mRNA、神经生长因子(NGF)/神经生长因子受体络氨酸激酶(trKa)的mRNA表达,Elisa方法检测肺组织中P物质(Sbstance P,SP)表达含量。结果:LPS组小鼠咳嗽次数及气道反应性明显高于空白组小鼠,提示LPS模型小鼠造模成功,并且祛风宣肺方能明显降低模型小鼠咳嗽次数、气道反应性和小鼠肺部炎症,相关数据具有统计学意义;祛风宣肺汤可以显著的减少LPS升高的TRPV1的表达和神经源性炎症,包括上游调控轴NGF/trKa的表达。结论:结合目前对于感染后气道高反应性的认识,考虑祛风宣肺汤可能是通过减少NGF/trKa来降低TRPV1的表达和神经源性炎症,并以此来降低气道高反应性。
Objective: To observe the therapeutic effect of Qufeng Xuanfei Decoction on cough reflex sensitivity and neurogenic inflammation in animal model of postin-fectious cough (PIC) induced by lipopolysaccharide (LPS) Mechanisms. Methods: Twenty-four male ICR mice were randomly divided into blank group, LPS group and TCM group by digital randomization method. Each group of 8, through the intraperitoneal injection of a certain concentration of LPS cause mice with post-infection cough airway hyperresponsiveness model, in which the Chinese medicine group and given Qufeng Xuanfei Tang gavage. The numbers of mice coughing after ammonia stimulation, the airway reactivity of pulmonary function test, the expression of transient receptor potential vanilloid 1 (TRPV1) Western Blot (WB) was used to detect the expression of capsaicin receptor TRPV1 protein in lung tissue. Real-time PCR was used to detect the expression of TRPV1 and substance P neuropeptide receptor 1 (NK1) MRNA, nerve growth factor (NGF) / nerve growth factor receptor tyrosine kinase (trKa) mRNA expression, Elisa method to detect the content of substance P (SPst SP, SP) in lung tissue. Results: The number of cough and airway responsiveness in LPS group were significantly higher than those in blank group, suggesting that LPS model mice succeeded in modeling, and Qufeng Xuanfei Decoction could significantly reduce the cough frequency, airway responsiveness and Mice lung inflammation, the relevant data with statistical significance; Qufeng Xuanfei decoction can significantly reduce LPS elevated TRPV1 expression and neurogenic inflammation, including the upstream regulatory axis of NGF / trKa expression. Conclusion: Combined with the current understanding of airway hyperresponsiveness after infection, expelling Fengfeng Xuanfei Decoction may reduce TRPV1 expression and neurogenic inflammation by decreasing NGF / trKa, and thus reduce airway hyperresponsiveness.
