以纯化的HSP27和HSP70通过WesternBlot检测接触不同浓度苯和苯中毒工人血浆中相应抗体水平。结果表明:苯中毒组HSP70抗体水平高于其它任一组,其它组间无显著性差异;而HSP27抗体水平无组间差异,提示HSP70抗体水平均高可能与苯及其代谢产物使细胞应激反应增强有关。P<0.01分别与各组比较。讨论热应激蛋白是原核生物和真核生物产生的具有高度保守性的蛋白家族。细胞在热、创伤、感染、重金属、化学物质、自由基和能量代谢抑制剂等多种因素作用下都被诱导产生热应激蛋白[3]。本实验结果表明,苯中毒组血浆HSP70抗体水平明显高于对照组。苯是明确的致癌剂,其对机体的慢性毒作用主要是通过中间代谢产物作用于造血系统。其重要的中间代谢产物主要有环氧化苯、氢醌和邻苯二酚[6]。我们检测发现苯中毒组的HSP70抗体水平明显高于其它组。HSPs抗体产生机制有两点与苯的毒作用机制一致,即抗原的变异和隐蔽抗原的释放及遗传因素所致[7]。苯及其代谢产物对细胞的直接刺激可使HSPs水平升高。另外,苯在代谢过程中还可产生一些自由基[8],自由基可诱导HSPs产生。有报道,暴露于苯的大鼠肝、骨髓及红细胞SOD活性高于对照组(P<?
Purified HSP27 and HSP70 were detected by Western Blot with different concentrations of benzene and benzene poisoning plasma levels of the corresponding antibodies. The results showed that the level of HSP70 antibody in benzene poisoning group was higher than that of any other group, but there was no significant difference in other groups. However, there was no significant difference between the two groups in the level of HSP27 antibody, suggesting that the high level of HSP70 antibody might be related to the stress of benzene and its metabolites Response enhancement related. The mechanisms of HSPs production are consistent with the toxic mechanism of benzene, which is the variation of antigens and the release of covert antigens and genetic factors [7]. Direct stimulation of cells with benzene and its metabolites can increase the level of HSPs.
