研究碱性成纤维细胞生长因子 (basicfibroblastgrowthfactor,bFGF)对自发性高血压大鼠 (SHR)和WKY大鼠血管一氧化氮 (NO)及内皮素生成的影响。取SHR和WKY大鼠主动脉制成血管薄片 ,以 10、10 0ng/mlbFGF(终浓度 )分别孵育 6h ,测定血管组织中一氧化氮合酶 (NOS)活性及孵育液中亚硝酸盐 (NO 2 )和内皮素含量。结果显示 :SHR主动脉组织NOS活性较WKY的低 17 6 % (P <0 0 1) ,血管薄片孵育液中NO 2 含量低 5 9 7% (P <0 0 1) ,内皮素浓度高 37 4% (P <0 0 1)。在低、高浓度bFGF刺激下 ,SHR主动脉组织NOS活性分别增高 2 9 7%和 5 9 6 %(均P <0 0 1) ,主动脉薄片孵育液中NO 2 含量分别增加 2 8 2 % (P <0 0 5 )和 70 5 % (P <0 0 1) ,主动脉内皮素的生成分别增加 2 4 1%和 44 5 % (均P <0 0 1) ;WKY大鼠主动脉组织NOS活性分别增高 2 4 4%和 5 3 7% (均P <0 0 1) ,主动脉薄片孵育液中NO 2 含量分别增加 18 8%和 2 5 9% (均P <0 0 1) ,主动脉内皮素的产生分别增加84 1%和 93 1% (均P <0 0 1)。结果表明 :bFGF对SHR和WKY大鼠主动脉NO及内皮素的产生具有调变作用。
To investigate the effect of basic fibroblast growth factor (bFGF) on nitric oxide (NO) and endothelin (ET) in spontaneously hypertensive rats (SHR) and WKY rats. Blood vessels were obtained from the aorta of SHR and WKY rats and incubated with 10, 10 ng / ml bFGF (final concentration) for 6 h respectively. The activities of nitric oxide synthase (NOS) and nitrite (NO 2) and endothelin content. The results showed that the activity of NOS in SHR aorta was 17 6% lower than that of WKY (P <0.01), NO 2 5 9 7% (P 0 01) was lower in vascular thin slice and the concentration of endothelin was 37 4% (P <0 0 1). Under low and high concentrations of bFGF stimulation, the activity of NOS in SHR aorta increased by 2 9 7% and 59 6%, respectively (all P 0 01), and the content of NO 2 in aorta slices increased by 28 2% (P <0.05) and 70.5% (P <0.01) respectively. The production of aortic endothelin increased by 24.1% and 44.5% respectively (all P <0.01). The aortic tissue of WKY rats NOS activity increased by 224% and 537%, respectively (all P <0.01), NO 2 content increased by 18.8% and 259% (P <0.01) in aorta sheet, Aortic endothelin production increased by 84 1% and 93 1%, respectively (all P <0.01). The results showed that: bFGF modulates NO and endothelin production in aortas of SHR and WKY rats.
