目的:制备茶藨子木层孔菌多糖硫酸酯,研究其体外的抗肿瘤活性,并与茶藨子木层孔菌多糖比较。方法:采用氯磺酸-甲酰胺法进行硫酸化,采用MTT法检测对人卵巢癌细胞SKOV-3和人乳腺癌细胞MDA231的影响。结果:制备了3种茶藨子木层孔菌多糖硫酸酯,硫取代度分别为1.48,1.83和2.23,其红外光谱中均具有SO和C-O-S的伸缩振动峰。茶藨子木层孔菌多糖对肿瘤细胞无明显的影响,而其硫酸酯可以抑制SKOV-3和MDA231细胞的生长。结论:成功制备了没有降解的3种硫酸酯,茶藨子木层孔菌多糖经硫酸化后对于肿瘤细胞的抑制作用增强。
OBJECTIVE: To prepare the polysaccharide sulphate from Pseudoterraceria przewalskii and study its anti-tumor activity in vitro. Methods: Sulfated by chlorosulfonic acid-formamide method, the effect of MTT assay on human ovarian cancer cell line SKOV-3 and human breast cancer cell line MDA231. Results: Three kinds of polysaccharides sulfate were prepared. Their sulfur substitution degrees were 1.48, 1.83 and 2.23, respectively. Their infrared spectra showed the stretching vibration peaks of SO and C-O-S. Scutellaria microporensis polysaccharides had no obvious effect on tumor cells, while sulfate inhibited the growth of SKOV-3 and MDA231 cells. CONCLUSION: Three kinds of sulfate esters without degradation were successfully prepared. The inhibitory effect of polysaccharides from Porphyra yezoensis on tumor cells was enhanced after sulfated.
