目的 重组腺病毒介导 HOSM基因转染胰腺癌细胞后 ,用 RT- PCR方法检测外源性 HOSM基因的表达情况。方法 构建了含人抑瘤素 M(OSM,oncostatin M)基因的复制缺陷型重组腺病毒载体 ad- HSOM,以 ad- HOSM转染胰腺癌细胞株后 ,提取细胞的总 RNA,用 RT- PCR法检测 HOSM基因在胰腺癌中的表达。结果 HOSM基因在转染细胞中有效的表达。结论 RT- PCR法能有效的检测外源性 HOSM基因在胰腺癌中的表达 ,为重组腺病毒介导 HOSM基因用于胰腺癌治疗的实验研究奠定了基础
Objective To detect the expression of exogenous HOSM gene by RT-PCR after the recombinant adenovirus-mediated HOSM gene was transfected into pancreatic cancer cells. Methods The recombinant adenovirus vector ad-HSOM containing human oncostatin M (OSM) gene was constructed and transfected into pancreatic cancer cell line ad-HOSM to extract the total RNA of the cell line. RT-PCR Method to detect the expression of HOSM gene in pancreatic cancer. Results The HOSM gene was effectively expressed in transfected cells. Conclusion The RT-PCR method can effectively detect the expression of exogenous HOSM gene in pancreatic cancer, which lays the foundation for the experimental study of recombinant adenovirus-mediated HOSM gene therapy in pancreatic cancer
