目的:探讨双氢青蒿素(dihydroartemisinin,DHA)对人前列腺癌细胞迁移和侵袭能力的影响及可能机制。方法:人前列腺癌细胞株PC-3与DU145经不同浓度(0、25、50、100μmol/L)的DHA处理48 h,运用细胞划痕和细胞侵袭实验检测细胞的迁移和侵袭能力。采用荧光定量PCR和Weston blot检测波形蛋白(Vimentin)和上皮-钙粘连素(E-Cadherin,CDH1)基因和蛋白表达水平。结果:不同浓度DHA均能显著抑制PC-3和DU145迁移和侵袭能力(P<0.001)。不同浓度DHA均能明显下调Vimentin m RNA和蛋白的表达(P<0.05),上调CDH1 m RNA和蛋白的表达(P<0.05)。结论:双氢青蒿素能显著抑制人前列腺癌细胞的迁移和侵袭能力,其可能机制是DHA干扰Vimentin表达及诱导E-Cadherin表达,且高浓度DHA可能存在其他途径抑制前列腺癌细胞的迁移和侵袭能力。
Objective: To investigate the effect of dihydroartemisinin (DHA) on the migration and invasion of human prostate cancer cells and its possible mechanism. METHODS: Human prostate cancer cell lines PC-3 and DU145 were treated with DHA at different concentrations (0, 25, 50 and 100 μmol / L) for 48 h. Cell migration and invasion were evaluated by cell scratch and cell invasion assay. Fluorescent quantitative PCR and Weston blot were used to detect the expression of vimentin and E-cadherin (CDH1) gene and protein. RESULTS: DHA at different concentrations significantly inhibited the migration and invasion of PC-3 and DU145 (P <0.001). DHA at different concentrations could significantly down-regulate the expression of Vimentin mRNA and protein (P <0.05) and up-regulate the expression of CDH1 mRNA and protein (P <0.05). CONCLUSION: Dihydroartemisinin can significantly inhibit the migration and invasion of human prostate cancer cells. The possible mechanism is that DHA interferes with Vimentin expression and induces E-Cadherin expression, and high concentrations of DHA may have other pathways to inhibit the migration and invasion of prostate cancer cells Invasive ability.
