补肾生血方剂对苯诱发再生障碍性贫血小鼠造血及免疫功能调节的研究

目的:通过观察补肾生血方剂对苯诱发再生障碍性贫血模型小鼠免疫及造血功能的影响,初步探讨中药的调节机制。方法:将清洁级Balb/c小鼠60只随机分为正常对照组、再障模型组和补肾生血方剂治疗组3组各20只;对照组注射玉米油4ml/kg;模型组和治疗组注射苯油混合液(苯剂量为1ml/kg体重),连注14天;然后治疗组小鼠按临床等效剂量的5倍灌胃给以相当于生药量为5.92g/(kg·d)的补肾生血方药,连续4周。结果:(1)模型组和治疗组小鼠体重及脾指数和胸腺指数较对照组均下降,差异有统计学意义(P<0.01);而治疗组小鼠胸腺指数和脾指数明显高于模型组,差异有统计学意义(P<0.01)。(2)模型组和治疗组小鼠外周血WBC、RBC、Hb、PLT的水平较对照组均明显下降,差异有统计学意义(P<0.01),且以模型组最明显;而治疗组与模型组比较,除RBC外,余3项指标均高,差异有统计学意义(P<0.01)。(3)模型组和治疗组骨髓组织较对照组均有不同程度的病理改变,且以模型组最明显,而治疗组的病理变化较模型组轻微。(4)模型组和治疗组小鼠血清sIL-2含量较对照组明显升高,差异有统计学意义(P<0.01);而TNF-α含量略升高,但差异无统计学意义(P>0.05)。治疗组sIL-2含量低于模型组,差异有统计学意义(P<0.01);但TNF-α含量较模型组无统计学差异(P>0.05)。结论:补肾生血方剂能明显改善苯诱发再障小鼠模型的骨髓造血功能,并可有效调节血清TNF-α、sIL-2的水平,部分纠正了模型小鼠免疫功能的紊乱。 OBJECTIVE: To observe the effect of Bushen Shengxue Prescription on immune and hematopoietic function in mice induced by benzene-induced aplastic anemia, and to explore the regulatory mechanism of traditional Chinese medicine. METHODS: 60 Balb/c mice of clean grade were randomly divided into normal control group, aplastic anemia model group, and 20 rats in each group. The control group was injected with corn oil 4 ml/kg; the model group and treatment group were injected. The benzene-oil mixture (benzene dosage was 1 ml/kg body weight) was injected for 14 days; then the mice in the treatment group were given 5 times the clinically equivalent dose and the equivalent amount of crude drug was 5.92 g/(kg·d). Bushenshengxue Recipe, for 4 weeks. Results: (1) The body weight, spleen index and thymus index in the model group and the treatment group were lower than those in the control group (P<0.01). The thymus index and spleen index in the treatment group were significantly higher than those in the control group. In the group, the difference was statistically significant (P<0.01). (2) The levels of WBC, RBC, Hb, and PLT in the peripheral blood of the model group and the treatment group were significantly lower than those in the control group (P<0.01), and the difference was significant in the model group; Compared with the model group, except for RBC, the remaining 3 indicators were high, and the difference was statistically significant (P<0.01). (3) Compared with the control group, the bone marrow tissue in the model group and the treatment group had different degrees of pathological changes, and the model group had the most obvious pathological changes, while the treatment group had slighter pathological changes than the model group. (4) The levels of sIL-2 in serum of the model group and the treatment group were significantly higher than those in the control group (P<0.01). However, the levels of TNF-α were slightly higher, but the difference was not statistically significant. >0.05). The content of sIL-2 in the treatment group was lower than that in the model group, and the difference was statistically significant (P<0.01). However, the content of TNF-α was not significantly different from the model group (P>0.05). Conclusion: Bushen Shengxue Prescription can significantly improve bone marrow hematopoiesis in benzene-induced aplastic anemia model, and can effectively regulate serum TNF-α and sIL-2 levels, partially correcting the disorder of immune function in model mice.
