目的初步探讨四氧嘧啶(Alloxan)所致成功高血糖模型的影响因素,为辅助降血糖功能检验方法的合理应用提供实验依据。方法选取清洁级昆明小鼠,采用腹腔注射Alloxan,每个剂量分2次注射,每次给予所设剂量的50%。按照研究目的,分成4个研究组。动物性别对模型的影响:选取雌性和雄性小鼠各27只,腹腔注射Alloxan200 mg/kg.bw;Alloxan剂量对模型的影响:小鼠120只,随机分为300、260、220和180 mg/kg.bw剂量组,每组30只;腹腔和尾静脉注射途径对模型的影响:小鼠60只分为2组,每组30只,一组腹腔注射Alloxan,分2次给药,每次130 mg/kg.bw,2次间隔24 h。另一组尾静脉注射Alloxan 80 mg/kg.bw;不同空腹血糖值的对模型的影响:整个试验周期,空腹血糖值在10 mmol/L~<15 mmol/L、15 mmol/L~<20 mmol/L、20 mmol/L~<25 mmo l/L、25 mmol/L~<30 mmol/L、≥30 mmol/L 5组小鼠对高血糖模型的影响。结果选择雌性昆明小鼠,采用260 mg/kg.bw腹腔注射方式(分2次给药),挑选空腹血糖值在20 mmol/L~<25 mmol/L的个体作为造模成功个体,可以获得一个相对稳定的高血糖模型。结论选择昆明种小鼠作为Alloxan糖尿病模型动物,动物的性别、Alloxan的剂量、个体空腹血糖值大小对模型的建立有影响。
Objective To investigate the influencing factors of successful hyperglycemia model induced by Alloxan and provide experimental basis for the reasonable application of the method of hypoglycemic function test. Methods Kunming mice of clean grade were selected. Alloxan was injected intraperitoneally. Each dose was injected in two times, and each dose was given 50% of the dose. According to the research purpose, divided into four research groups. Effects of animal sex on the model: 27 female and male mice were selected and Alloxan was injected intraperitoneally at a dose of 200 mg / kg.bw. The effect of Alloxan dosage on the model: 120 mice were randomly divided into 300, 260, 220 and 180 mg / kg.bw dose group, 30 rats in each group. Effects of intraperitoneal and tail vein injection on the model: 60 mice were divided into two groups of 30 rats in each group. Alloxan was intraperitoneally injected in one group, 130 mg / kg.bw, two intervals of 24 h. Another group of tail vein injection of Alloxan 80 mg / kg.bw; different fasting blood glucose levels on the model: the entire test period, fasting blood glucose values of 10 mmol / L ~ <15 mmol / L, 15 mmol / L ~ <20 The effects of 20 mmol / L ~ 25 mmol / L, 25 mmol / L ~ 30 mmol / L, and 30 mmol / L 5 on the hyperglycemic model mice were investigated. Results Female Kunming mice were selected by intraperitoneal injection of 260 mg / kg.bw (twice), and those individuals whose fasting blood glucose level was between 20 mmol / L and 25 mmol / L were selected as successful individuals A relatively stable model of hyperglycemia. Conclusion Kunming mice were selected as model animals for Alloxan diabetes. The sex of animals, the dose of Alloxan and the fasting blood glucose level of individuals affected the model establishment.
