本文讨论药物通过鼻腔吸收而发挥全身作用的给药系统,这类药物主要包括:口服后个体差异大而生物利用度低者,如心得安等,及口服后易被胃肠道破坏或不被吸收者,如黄体酮、胰岛素等激素、多肽与蛋白质类药物。在设计鼻腔给药系统时应考虑鼻粘膜的绒毛运动及选用适宜的吸收促进剂,以增加药物被鼻腔的吸收。本文简要地阐述了影响鼻腔吸收的诸因素,如药物本身的理化性质,鼻粘膜的生理、病理状况以及给药系统的处方和给药方式。
