目的 :观察半相合骨髓移植对 GVHD或 GVL的影响。方法 :以 C57BL - 6小鼠急性放射病为模型 ,进行半相合小鼠骨髓移植 ,同时设异基因骨髓移植为对照 ,观察移植后小鼠存活率、造血重建、GVHD或 GVL作用。结果 :GVHD中 ,半相合骨髓移植组 30天死亡率低于基因骨髓移植。半相合骨髓移植与异基因骨髓移植比较 ,外周血象或骨髓 CFU - GM恢复快 ,以 9天以后明显 ,脾结节数及嵌合体测定高于异基因组。体内混合淋巴细胞反应证实半相合骨髓移植较异基因骨髓移植 GVHD小。在 GVL中 ,异基因骨髓移植组主要死于 GVHD。半相合骨髓移植组 30天生存率仍为 5 6 .3% ,表现出明显 GVL作用。结论 :半相合骨髓移植在减轻 GVHD同时 ,产生明显的 GVL作用。
Objective: To observe the influence of hemizygous bone marrow transplantation on GVHD or GVL. Methods: Acute radiation sickness of C57BL - 6 mice was used as a model to carry out bone marrow transplantation in semi - compatible mice. At the same time, allogeneic bone marrow transplantation was used as a control to observe the survival rate, hematopoietic reconstitution, GVHD or GVL in transplanted mice. Results: In GVHD, the 30-day mortality rate in haploidentical bone marrow transplantation group was lower than that in gene marrow transplantation. Compared with allogeneic bone marrow transplantation, hematopoietic recovery of CFU - GM in peripheral blood or bone marrow was faster in haploidentical bone marrow transplantation than in allogeneic bone marrow transplantation. In vivo mixed lymphocyte reaction confirmed semi-matched bone marrow transplantation than allogeneic bone marrow transplantation GVHD small. In GVL, the allogeneic bone marrow transplant group mainly died of GVHD. The 30-day survival rate in the haploidentical bone marrow transplantation group was still 56.3%, showing obvious GVL effect. CONCLUSION: Hemiadjuvant bone marrow transplantation can reduce GVHD and produce obvious GVL.
