目的:观察BAG3蛋白在胶质瘤中的表达,及其ser187位点磷酸化与胶质瘤间的相关性。方法:收集胶质瘤组织标本,进行分子生物学实验,观察BAG3蛋白在胶质瘤组织中的表达情况及BAG3蛋白ser187位点磷酸化对细胞侵袭的影响。结果:(1)IHC:BAG3在Ⅳ级胶质瘤组织表达显著高于正常脑组织及Ⅰ、Ⅱ级胶质瘤组织(P<0.05)。(2)Western Blot:BAG3蛋白在Ⅱ、Ⅲ、Ⅳ级别胶质瘤表达量,显著高于正常脑组织(P<0.05),且相对于Ⅱ级胶质瘤,在Ⅲ、Ⅳ级胶质瘤中表达显著增高(P<0.05),而在Ⅲ级、Ⅳ级胶质瘤组织中表达量之间无显著性差异(P>0.05)。(3)三维Matrigel transwell转移小室实验,S187A-BAG3蛋白过表达U87细胞侵袭Matrigel模拟基质膜的细胞数显著高于U87细胞和S187D-BAG3过表达U87细胞。结论:BAG3蛋白胶质瘤组织中表达量高,且在Ⅳ级胶质瘤组织中,BAG3蛋白的表达明显高于Ⅰ、Ⅱ级及正常脑组织,且S187位点磷酸化能抑制U87细胞的侵袭。
OBJECTIVE: To observe the expression of BAG3 protein in gliomas and the relationship between its ser187 phosphorylation and glioma. Methods: The glioma tissue samples were collected for molecular biology experiments to observe the BAG3 protein expression in glioma tissue and BAG3 protein ser187 site phosphorylation on cell invasion. Results: (1) The expression of IHC: BAG3 in grade Ⅳ gliomas was significantly higher than that in normal brain tissues and grade Ⅰ and grade Ⅱ gliomas (P <0.05). (2) Western Blot: The expression of BAG3 protein in Grade II, III and IV gliomas was significantly higher than that in normal brain tissues (P <0.05). Compared with Grade II Gliomas, (P <0.05), while there was no significant difference in grade Ⅲ and Ⅳ glioma (P> 0.05). (3) Three-dimensional Matrigel transwell transfer chamber experiments showed that the number of S187A-BAG3 overexpressing U87 cells infiltrating Matrigel mimic stromal membranes was significantly higher than that of U87 cells and S187D-BAG3 overexpression U87 cells. CONCLUSION: BAG3 protein is highly expressed in glioma tissues, and in grade IV gliomas, the expression of BAG3 protein is significantly higher than that in grade Ⅰ, Ⅱ and normal brain tissues, and S187 phosphorylation can inhibit the expression of U87 cells Invasion.
