目的 探讨人胃癌中转录信号传导子和激活子 3(STAT3)的表达情况 ,分析其表达与胃癌临床病理因素的关系。方法 应用固定化蛋白质印迹法 (Westernblot)检测 42例胃癌组织、转移淋巴结及邻近的正常胃粘膜中STAT3蛋白的表达。结果 胃癌组织中STAT3蛋白表达水平明显高于正常胃粘膜 ,平均为正常胃粘膜中的 2 .14倍 (P <0 .0 5 ) ;低分化胃癌STAT3蛋白表达水平高于高分化胃癌 [分别为正常胃粘膜的 ( 2 .44± 2 .99)倍和 ( 1.0 3± 0 .88)倍 ,P <0 .0 5 ] ;Ⅲ、IV期胃癌STAT3蛋白表达水平高于I、II期胃癌 [分别为正常胃粘膜的 ( 2 .5 9± 3.2 2 )倍和 ( 1.2 4±0 .92 )倍 ,P <0 .0 5 ] ;有淋巴结转移的胃癌STAT3蛋白表达水平高于无淋巴结转移胃癌 [分别为正常胃粘膜的 ( 2 .5 5± 3.12 )倍和 ( 1.11± 0 .82 )倍 ,P <0 .0 5 ] ;转移淋巴结STAT3蛋白表达水平高于阴性淋巴结。结论 STAT3过表达可能在胃癌的发生、发展过程中发挥重要作用 ,其表达与胃癌组织学分化程度、TNM分期及淋巴结转移有关。
Objective To investigate the expression of STAT3 in human gastric cancer and analyze the relationship between its expression and clinicopathological factors of gastric cancer. Methods The expression of STAT3 protein was detected in 42 gastric cancer tissues, metastatic lymph nodes and adjacent normal gastric mucosa by immobilized Western blotting. Results The expression of STAT3 protein in gastric cancer tissues was significantly higher than that in normal gastric mucosa, with an average of 2.14 times (P < 0.05) in normal gastric mucosa. The expression of STAT3 protein in poorly differentiated gastric cancer was higher than that in well-differentiated gastric cancer [respectively The normal gastric mucosa was (2.44± 2.99) folds and (1.0 3±0.88) folds, P<0.05. The expression of STAT3 protein in stage III and IV gastric cancer was higher than that in stage I and II gastric cancer [ The normal gastric mucosa were (2.59±3.22) folds and (1.24±0.92) folds, P<0.05; the expression of STAT3 protein in gastric cancer with lymph node metastasis was higher than that in no gastric cancer without lymph node metastasis. [(2.5 ± 3.12)-fold and (1.11 ± 0.82)-fold for normal gastric mucosa, respectively, P <0.05); metastatic lymph node STAT3 protein expression level was higher than negative lymph node. Conclusion Overexpression of STAT3 may play an important role in the occurrence and development of gastric cancer. The expression of STAT3 may be related to histological differentiation, TNM stage and lymph node metastasis.
