目的 探讨流式细胞术CD45辅助设门在白血病表型分析中的作用。方法 根据CD45在所有白细胞上都有表达 ,但在不同系成熟细胞或同系不同发育阶段细胞表达量不同的特点 ,在流式细胞仪上采用对数CD45 -线性侧向角光散射 (SSC)图设门及前向角光散射 (FSC) -侧向角光散射 (SSC)图设门进行免疫表型分析。结果 用FSC -SSC表示则白血病细胞群与正常淋巴细胞群分布领域的境界不清 ,而用CD45 -SSC图表示可把两群细胞明显区分开来 ,且两种设门方法所得表型结果有不同之处。结论 CD45 -SSC图设门提高了白血病免疫学分型的准确性 ,尤其对白血病细胞比例较低患者提供了一个可靠的方法学手段 ,且该方法易于掌握 ,无主观因素 ,重复性好。
Objective To investigate the role of flow cytometry CD45-assisted docking in leukemia phenotype analysis. Methods According to the expression of CD45 on all leukocytes, but different cell lines at different developmental stages in different lines, CD45 - linear lateral light scattering (SSC) images were acquired by flow cytometry Gate and Forward Angle Light Scattering (FSC) - Lateral Angle Light Scattering (SSC) maps were set up for immunophenotyping. Results The FSC-SCC indicates that the realm of leukemia and normal lymphocyte population distribution is ill-defined, whereas the CD45-SSC indicates that the two groups of cells are clearly distinguishable and that the phenotypic results obtained by the two portal methods are the difference. Conclusion The CD45-SSC map provides an accurate method to improve the immunological typing of leukemia. It provides a reliable methodological method for leukemia patients with low proportion of leukemia cells. The method is easy to master, subjective and reproducible.
