目的研究川芎嗪体外对活化T细胞的作用及机制。方法应用植物血凝素(PHA)刺激正常人外周血T细胞,经川芎嗪处理后,应用四甲基偶氮唑盐(MTT)微量酶反应比色法检测细胞增殖,流式细胞术检测CD4+和CD8+T细胞上CD25(白细胞介素-2受体,IL-2R)及负性协同刺激分子程序性死亡分子1(PD-1)的表达,ELISA法检测细胞因子IL-2水平变化。结果川芎嗪(150μg/ml)处理72 h后,能明显抑制T细胞的增殖和活化(P<0.01),并降低IL-2水平(P<0.01);能抑制CD4+和CD8+T细胞上IL-2R的表达(P<0.05);对负性协同刺激分子PD-1的表达作用不明显(P>0.05)。结论川芎嗪对免疫过激T细胞具有一定的抑制作用,其对T细胞的免疫抑制作用可能与降低IL-2的分泌和抑制IL-2R(CD25)的表达有关。
Objective To study the effect and mechanism of ligustrazine on activated T cells in vitro. METHODS: Phytohemagglutinin (PHA) was used to stimulate normal human peripheral blood T cells. After treated with tetramethylpyrazine, cell proliferation was detected by MTT colorimetric enzyme reaction colorimetric assay, and CD4+ was detected by flow cytometry. CD25 (interleukin-2 receptor, IL-2R) and CD8+ T cells were co-stimulated with the expression of programmed cell death 1 (PD-1), and IL-2 levels were measured by ELISA. RESULTS: After treated with tetramethylpyrazine (150μg/ml) for 72 hours, T cell proliferation and activation were significantly inhibited (P<0.01), IL-2 level was decreased (P<0.01), and CD4+ and CD8+ T cells were inhibited. The expression of -2R (P<0.05); The expression of negative co-stimulatory molecule PD-1 was not significant (P>0.05). Conclusion Ligustrazine has a certain inhibitory effect on immune-stimulated T cells, and its immunosuppressive effect on T cells may be related to the decrease of IL-2 secretion and the inhibition of IL-2R (CD25) expression.
