目的利用计算机软件预测人感染H7N9禽流感病毒血凝素(HA)和神经氨酸酶(NA)的B细胞表位。方法以HA、NA的氨基酸序列为基础,用Protean、PORTER、MEGA等生物信息学软件综合预测HA、NA蛋白可能的B细胞表位,再结合吴玉章的平均抗原指数方案进一步筛选。结果 HA蛋白最有可能的B细胞表位位于其N端的第53~57、179~183区段,HA蛋白的其它区段:62~66、98~101、164~166、315~320、424~426、443~445、493~496等也有可能存在B细胞表位。NA蛋白最有可能的B细胞表位位于N端的第321~329、379~386区段,NA蛋白的其他区段:46~48、52~54、74~78、136~141、145~150、160~164、194~197、246~248、333~343、426~431等也有可能存在B细胞表位。结论综合多种方案预测了H7N9病毒HA、NA蛋白的B细胞表位,为人感染H7N9禽流感病毒表位疫苗研究提供了实验基础。
Objective To predict the B cell epitopes of human haemagglutinin (HA) and neuraminidase (NA) in H7N9 bird by using computer software. Methods Based on the amino acid sequences of HA and NA, the potential B cell epitopes of HA and NA proteins were predicted by bioinformatics software such as Protean, PORTER and MEGA, and then further screened with the average antigen index of Wu Yuzhang. Results The most probable B cell epitope of HA protein was located in the 53 ~ 57,179 ~ 183 segment of its N terminal, the other segments of HA protein: 62 ~ 66,98 ~ 101,164 ~ 166,315 ~ 320,424 ~ 426,443 ~ 445,493 ~ 496, etc. There may be B cell epitopes. The most probable B cell epitopes of NA protein are located in the N-terminal region from 321 to 329, 379 to 386 and the other NA segments are from 46 to 48, 52 to 54, 74 to 78, 136 to 141, 145 to 150 , 160 ~ 164,194 ~ 197,246 ~ 248,333 ~ 343,426 ~ 431, etc. There may be B cell epitopes. Conclusions A variety of protocols are used to predict the B cell epitopes of HA and NA proteins of H7N9 virus and provide the experimental basis for the study of human H7N9 avian influenza virus epitope vaccine.
