目的:通过建立Balb/c小鼠CT26人工种植模型,研究复方藤梨根制剂对肿瘤抑制效果及对MVD表达的影响。方法:①100只7周龄雌雄各半Balb/c小鼠随机分为5组,即空白对照组、荷瘤对照组、低剂量给药组、中剂量给药组、高剂量给药组。各组分别皮下接种造模。②造模后第2 d开始连续灌胃3周。给药组予低浓度0.5 g/mL、中浓度1 g/mL、高浓度2 g/mL复方藤梨根制剂灌胃,空白对照组、荷瘤对照组每天以生理盐水灌胃。③所有小鼠第22 d处死,观察各组的瘤重、抑瘤率,免疫组化检测各组小鼠肿瘤CD34标记的MVD。结果:FFTLG低、中、高剂量组抑瘤率分别为-7.05%、23.03%、32.91%;中、高剂量给药组的瘤重与低剂量组和荷瘤组有明显差异(P<0.05),中、高剂量给药组的MVD对比荷瘤对照组有显著性差异(P<0.05)。结论:复方藤梨根制剂能抑制肿瘤生长,减少CT26瘤体中MVD,具有抗肿瘤血管生成作用。
OBJECTIVE: To study the inhibitory effect of Fufang Tengligen on tumor and its effect on the expression of MVD in Balb / c mouse CT26 model. Methods: ①100 male and female Balb / c mice, 7 weeks old, were randomly divided into 5 groups: blank control group, tumor-bearing control group, low dose group, middle dose group and high dose group. Each group were subcutaneously inoculated modeling. ② ② 2 d after the beginning of continuous gavage for 3 weeks. The rats in the treatment group were given gavage at a low concentration of 0.5 g / mL, medium concentration of 1 g / mL and high concentration of 2 g / mL. The blank control group and the tumor-bearing control group were intragastrically administered with normal saline. (3) All mice were sacrificed on the 22nd day, and the tumor weight and tumor inhibition rate were observed in each group. The tumor CD34-labeled MVD in each group was detected by immunohistochemistry. Results: The tumor inhibition rates of FFTLG low-dose, medium-dose and high-dose groups were -7.05%, 23.03% and 32.91% respectively. There was significant difference between medium and high dose groups in tumor weight and low dose group and tumor bearing group (P <0.05 ). The MVD of medium dose group and high dose group were significantly different from that of the control group (P <0.05). CONCLUSION: Fufang Tengligen can inhibit tumor growth, reduce MVD in CT26 tumor and have anti-tumor angiogenesis effect.
