目的研究白细胞介素-17(IL-17)对肝星状细胞的增殖及其对细胞周期变化的影响。方法以体外培养的肝星状细胞为研究对象,分别以200、400、600、800、1 000μg/L(终质量浓度)的IL-17刺激,并设空白对照组,分别培养24、48、72 h。用细胞计数法观察细胞的生长情况,MTT法检测IL-17刺激后肝星状细胞的增殖,流式细胞术检测不同浓度的IL-17对肝星状细胞增殖周期的影响。结果 IL-17对细胞的刺激增殖作用在一定的程度和范围内呈现剂量和时间依赖性,200μg/L的IL-17对细胞无明显的刺激作用(P>0.05),800μg/L的IL-17在细胞培养72 h后对细胞的刺激增殖作用最明显(P<0.01),1 000μg/L的IL-17对细胞则有抑制作用(P<0.01)。IL-17刺激肝星状细胞后能促使细胞由G0/G1期进入S期,细胞增殖以S期细胞比例为主。在一定范围内,细胞增殖指数随着IL-17浓度的增长而增加,而1 000μg/L的IL-17则抑制细胞周期,使细胞周期停滞于G1期(P<0.01)。结论 IL-17能刺激肝星状细胞的增殖,在一定的程度和范围内呈现剂量和时间依赖性,IL-17能够促进肝星状细胞由G0/G1期进入S期。
Objective To investigate the effects of interleukin-17 (IL-17) on the proliferation of hepatic stellate cells and its effect on the cell cycle. Methods Hepatic stellate cells cultured in vitro were stimulated with 200, 400, 600, 800, 1 000 μg / L (final concentration) of IL-17 and blank control group were cultured 24, 48, 72 h. The cell growth was observed by cell counting method. The proliferation of hepatic stellate cells was detected by MTT assay. The proliferation of hepatic stellate cells was detected by flow cytometry. Results IL-17 stimulated proliferation and proliferation in a dose and time-dependent manner to a certain extent and range. IL-17 at 200 μg / L had no significant effect on cells (P> 0.05). IL- 17 had the strongest effect on cell proliferation 72 h after cell culture (P <0.01), while IL-17 at 1 000 μg / L had an inhibitory effect on cells (P <0.01). Stimulation of hepatic stellate cells by IL-17 can induce cells to enter S phase from G0 / G1 phase, and cell proliferation mainly takes the proportion of S phase cells. Within a certain range, the cell proliferation index increased with the increase of IL-17 concentration, while 1 000 μg / L IL-17 inhibited the cell cycle and arrested the cell cycle in G1 phase (P <0.01). Conclusion IL-17 can stimulate the proliferation of hepatic stellate cells in a dose and time-dependent manner to a certain extent and extent. IL-17 can promote hepatic stellate cells to enter S phase from G0 / G1 phase.
