目的:探讨天舒胶囊对大鼠局灶性脑缺血再灌注损伤的保护作用,并初步探讨其作用机制。方法:雄性SD大鼠80只,随机分成假手术组、模型组、天舒胶囊低剂量组(0.6 g·kg-1)、天舒胶囊高剂量组(1.2 g·kg-1)。口服给药5 d后采用线栓法制作大鼠大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)90 min再灌注24 h模型。观察天舒胶囊对脑缺血再灌注大鼠神经缺失症状、脑组织含水量、脑梗死体积及脑组织病理形态学的影响。检测各组大鼠神经细胞凋亡和血清磷酸肌酸激酶脑型同工酶(CK-BB)水平。结果:与模型组比较,天舒胶囊明显降低脑梗死体积(低、高剂量脑梗死体积分别为29.11%,27.85%);血清CK-BB水平(低、高剂量分别为9.95,9.87 U·L-1)也明显降低(P<0.01),神经缺失症状和脑组织病理损害,神经细胞凋亡减少(P<0.01),脑组织含水量(低、高剂量分别为79.03%,78.89%)降低(P<0.05)。结论:天舒胶囊对大鼠脑缺血再灌注损伤具有保护作用,其通过降低血清CK-BB水平、减少神经细胞凋亡,保护脑缺血再灌注损伤。
Objective: To investigate the protective effect of Tianshu capsule on focal cerebral ischemia-reperfusion injury in rats and its mechanism of action. Methods: Eighty male Sprague-Dawley rats were randomly divided into sham operation group, model group, Tianshu capsule low dose group (0.6 g · kg -1) and Tianshu capsule high dose group (1.2 g · kg -1). After oral administration for 5 days, MCAO rats were reperfused for 24 hours by thread occlusion. To observe the effect of Tianshu capsule on neurological deficit symptoms, brain water content, volume of cerebral infarction and histopathology in rats with cerebral ischemia / reperfusion. Apoptosis of neurons and serum CK-BB level were detected in each group. Results: Compared with the model group, Tianshu capsule significantly reduced the volume of cerebral infarction (the volume of low and high dose cerebral infarction were 29.11% and 27.85% respectively); the serum CK-BB level (low and high dose were 9.95 and 9.87 U · L -1) were also significantly decreased (P <0.01). The neurological deficits and pathological changes of brain tissue and the apoptosis of nerve cells were decreased (P <0.01), and the water content of brain tissue (79.03% and 78.89% (P <0.05). CONCLUSION: Tianshu capsule has a protective effect on cerebral ischemia-reperfusion injury in rats. It protects cerebral ischemia-reperfusion injury by decreasing serum CK-BB level, decreasing neuronal apoptosis.
