目的:探讨caspase-3在慢性粒细胞白血病信号转导中的作用。方法:用脂质体转染法将bcr3/abl2ASO导入K562细胞中,通过检测细胞凋亡、caspase-3表达和caspase-3活性的变化,观察caspase-3在慢性粒细胞白血病信号转导中的作用。结果:bcr3/abl2ASO可诱导K562细胞凋亡;正常情况下caspase-3在K562细胞中有一定量的基础表达和活性,分别为(1.13±0.1)和(0.025±0.001);而K562发生凋亡时,caspase-3水平明显升高,其表达和活性分别为(2.93±0.6)和(0.903±0.003)。结论:caspase-3通过介导凋亡信号而在慢性粒细胞白血病信号转导中起重要作用。 Objective: To investigate the role of caspase-3 in signal transduction of chronic myelogenous leukemia. Methods: bcr3 / abl2ASO was transfected into K562 cells by lipofectamine 2000. The apoptosis, the expression of caspase-3 and the activity of caspase-3 were detected by flow cytometry. The expression of caspase-3 in the signal transduction of chronic myeloid leukemia effect. Results: bcr3 / abl2ASO induced the apoptosis of K562 cells. Under normal conditions, caspase-3 had a certain amount of basic expression and activity in K562 cells, which were (1.13 ± 0.1) and (0.025 ± 0.001) , caspase-3 levels were significantly increased, the expression and activity were (2.93 ± 0.6) and (0.903 ± 0.003). Conclusion: Caspase-3 plays an important role in the signal transduction of chronic myelogenous leukemia through mediating apoptosis signaling.
