目的:比较糙叶败酱提取物糙叶败酱粗多糖、糙叶败酱总木脂素、糙叶败酱总皂苷的体外抗肿瘤活性,初步筛选糙叶败酱抗癌作用有效成分。方法:采用四甲基偶氮唑盐比色法(monotetrazolium test,MTT)测定不同浓度糙叶败酱粗多糖、糙叶败酱总木脂素、糙叶败酱总皂苷分别对人肺源腺癌细胞SPCA-1、人肝癌细胞系HepG2、和人慢性髓系白血病细胞K562生长的影响。结果:糙叶败酱总木脂素、总皂苷对体外培养的肿瘤细胞SPCA-1、HepG2、K562均有抑制作用,且抑制作用与药物浓度及作用时间有相关性;糙叶败酱总木脂素对K562细胞生长的抑制作用尤为显著,400μg/ml,作用96h,IC50为21.16μg/ml;糙叶败酱粗多糖对SPCA-1细胞的增殖有抑制作用。结论:糙叶败酱不同成分有不同的抗肿瘤作用,糙叶败酱总木脂素对人慢性髓系白血病细胞生长有明显的抑制作用。
OBJECTIVE: To compare the antitumor activity of the extract of Patrinia prochterica leaf extract with crude polysaccharide, total lignans of Patrinia proctotypes and total saponins of Patrinia proctotype so as to screen the anti-cancer active ingredients of Patrinia moutain. Methods: MTT assay was used to determine the content of crude polysaccharides in Caulis Caevum, total lignans in Carapax putida, total saponin in Caulis Caragana, Cancer cell SPCA-1, human hepatoma cell line HepG2, and human chronic myeloid leukemia cell K562. Results: Total lignans and total saponins of Patrinia Lusitapi had inhibitory effects on the SPCA-1, HepG2 and K562 cells cultured in vitro, and the inhibitory effect was related to the drug concentration and time. The inhibitory effect of lipids on K562 cells was particularly significant, 400μg / ml, 96h, IC50 of 21.16μg / ml. The crude polysaccharide of Paeonia scabra inhibited the proliferation of SPCA-1 cells. CONCLUSION: The different components of Patrinia sinica have different antitumor effects. Total lignans of Patrinia prochora have a significant inhibitory effect on the growth of human chronic myeloid leukemia cells.
