The minimal model of marker glucose occlusion allows one to measure important parameters in human metabolism through a labeled IVGTT (Deep Vein Glucose Endurance Test). It usually takes 30 blood samples to measure the time and process of insulin and marker glucose accumulation, which provides the data basis for model recognition. In this article, we explore the issue of reducing the number of times a blood sample is extracted without compromising the quality of the parameter estimates. By applying the successive approximation approach and following a large number of best experimental design theories, we have obtained a reliable procedure to reduce the number of samples for a total of only 14 blood samples.
