目的 观察腰痛舒胶囊对一氧化氮在体外诱导的兔软骨细胞凋亡的影响。方法 体外培养的兔软骨细胞 ,分别加入硝普钠 (SNP)培养 12、2 4、4 8h ,用透射电镜和流式细胞仪观察软骨细胞凋亡的变化 ,并以不同剂量腰痛舒胶囊含药血清加入NO诱导的软骨细胞凋亡体系培养 2 4h ,采用annexinV PI方法检测凋亡情况。结果 在体外培养兔软骨细胞加入硝普钠 2 4h ,流式细胞仪检测凋亡率为 (74 0 5± 6 6 8) % ,对照组为 (3 15± 0 76 ) % (P <0 0 1) ;透射电镜可见较典型凋亡细胞形态改变 ,加入不同剂量腰痛舒胶囊含药血清 ,其凋亡率明显下降 ,并以高剂量最为显著。结论 硝普钠在体外能诱导软骨细胞凋亡 ;高剂量腰痛舒含药血清能明显降低NO诱导的软骨细胞凋亡 ,提示腰痛舒胶囊能抑制软骨细胞凋亡。
Objective To observe the effect of Yaotongshu capsule on the apoptosis of rabbit chondrocytes induced by nitric oxide in vitro. Methods Rabbit chondrocytes cultured in vitro were added with sodium nitroprusside (SNP) for 12, 24 and 48 hours respectively. Transmission electron microscopy and flow cytometry were used to observe the changes of chondrocyte apoptosis, and different doses of Yaotongshu capsule contained the drug. Nitric oxide-induced chondrocyte apoptosis was added to serum for 24 hours and the apoptosis was detected by annexin V PI method. Results Sodium nitroprusside was added to rabbit chondrocytes in vitro for 24 hours. The apoptosis rate was (74 0 5 ± 6 6) % in flow cytometry and (3 15 ± 0 76) % in control group (P <0 0. 1) The morphological changes of typical apoptotic cells were observed by transmission electron microscopy. Adding different doses of Yaotongshu capsule-containing serum, the apoptotic rate was significantly decreased, and the highest dose was the most significant. Conclusion Sodium nitroprusside can induce apoptosis of chondrocytes in vitro. High dose of Yaotongshu serum can significantly reduce NO-induced apoptosis of chondrocytes, suggesting that Yaotongshu capsule can inhibit chondrocyte apoptosis.
