目的 观察成骨生长肽 ( Osteogenic growth peptide,简称 OGP)对 SD大鼠鼠胚颅盖骨成骨细胞样细胞酶活性的影响。方法 无菌条件下培养鼠胚颅盖骨成骨细胞样细胞 ,分为不加 OGP的对照组和加 OGP的用药组 ,用药组在 10 -12 M~ 10 -8M OGP浓度范围内 ,再分为三组 (分别为 OGP浓度10 -12 M、10 -10 M、10 -8M) ,收集以上四组细胞及细胞培养液 ,测定它们的碱性磷酸酶及酸性磷酸酶活性。结果 用药组的碱性磷酸酶活性均显著地高于对照组 ( P<0 .0 1~ 0 .0 5,用药组 OGP10 -8M浓度、培养7天的细胞培养液中碱性磷酸酶活性为 0 .82± 0 .16U/ L,而对照组同样条件活性为 0 .2 3± 0 .10 U/ L) ,且随着 OGP浓度的升高碱性磷酸酶活性也升高 ,而酸性磷酸酶用药组与对照组无显著差异。结论 OGP能促进成骨细胞样细胞分泌碱性磷酸酶
Objective To observe the effect of osteogenic growth peptide (OGP) on the cellular activity of callus in osteoblasts of SD rats. Methods Callus osteoblast-like cells were cultured under aseptic conditions and divided into three groups: OGP-free control group and OGP-treated group. The drug group was subdivided into 10 -12 M ~ 10 -8 M OGP concentration range The three groups (OGP concentration 10-12 M, 10 -10 M, 10 -8 M respectively) were collected and the above four groups of cells and cell culture medium were collected to measure their alkaline phosphatase and acid phosphatase activities. Results Alkaline phosphatase activity in the drug-treated group was significantly higher than that in the control group (P <0.01-0.05, OGP10-8M concentration in the drug-treated group, alkaline phosphatase activity in the cell culture solution for 7 days 0.82 ± 0.16 U / L, while the control group had the same condition activity of 0.23 ± 0.10 U / L), and the activity of alkaline phosphatase increased with the increase of OGP concentration. However, Enzyme-treated group and control group no significant difference. Conclusion OGP can promote osteoblast-like cells to secrete alkaline phosphatase
