目的:研究健脾益智法对阿尔茨海默病(AD)大鼠海马区脑肠肽神经肽Y(NPY)m RNA表达水平的影响及其作用机制。方法:采用右侧海马注射凝聚态Aβ1-40的方法制作AD大鼠模型,以健脾益智混悬液、吡拉西坦片、0.9%氯化钠溶液分组灌胃给药连续治疗4周,采用半定量RT-PCR法检测比较各组大鼠海马区脑肠肽NPY m RNA的表达水平。结果:行为学评价分析得出,与模型对照组比较,中药组可以缩短Morris水迷宫实验逃避潜伏期,增加跨越平台次数。中药组AD大鼠海马区脑肠肽NPY m RNA表达水平明显高于模型对照组和西药组。结论:健脾益智法可以上调大鼠海马区脑肠肽NPY m RNA的表达水平,且上调作用明显优于西药组,对AD大鼠的学习记忆能力有明显的调节作用,为进一步研究及临床应用中医药防治AD提供实验依据。
Objective: To investigate the effect and mechanism of “Jianpi Yizhi” on neuropeptide Y (NPY) mRNA expression in hippocampus of Alzheimer’s disease (AD) rats. Methods: AD rat models were made by injection of condensed Aβ1-40 in the right hippocampus. The rats in the AD model were treated with spleen-qi suspension, piracetam tablets and 0.9% sodium chloride solution for continuous treatment for 4 weeks The expression of NPY m RNA in hippocampus of rats in each group was detected by semi-quantitative RT-PCR. Results: Behavioral evaluation analysis showed that, compared with the model control group, the traditional Chinese medicine group can shorten the escape latency of Morris water maze test and increase the number of cross platform. The expression level of brain natriuretic peptide NPY m RNA in hippocampus of AD rats was significantly higher than that in model control group and western medicine group. Conclusion: Jianpi Yizhi Decoction can upregulate the expression of brain natriuretic peptide NPY m RNA in hippocampus of rats, and its up-regulation effect is obviously better than that of Western medicine group. It has a significant regulatory effect on the learning and memory ability of AD rats. For further study and Clinical application of traditional Chinese medicine to provide experimental evidence AD.
