本实验采用钙离子细胞化学定位法、膜通透性细胞化学示踪法结合普通超微结构及EDX电镜分析,从细胞水平观察了肾近端小管上皮细胞缺血损伤情况以及药物对它的影响。实验结果表明:肌苷、川芎对细胞缺血损伤有保护作用。缺血期,细胞中及线粒体内钙离子增多。此时,质膜的损伤并不严重。缺血后复流,随着复流时间的延长,细胞中及线粒体内钙离子进行性增多,质膜的损伤也越来越重。线粒体内钙离子含量与胞质中钙离子含量有密切关系。
In this study, calcium ion cytochemical localization method, membrane permeability cytochemical tracing method combined with ordinary ultrastructure and EDX electron microscopy analysis of renal proximal tubular epithelial cells from the cellular level of ischemic injury and the impact of drugs on it . The experimental results show that inosine and ligusticum chuanxiong have a protective effect on cell ischemia injury. Ischemic period, cells and mitochondrial calcium increased. At this point, the plasma membrane damage is not serious. Reperfusion after ischemia, with the prolongation of reflow time, cells and mitochondrial calcium increase, the plasma membrane damage is more and more heavy. Mitochondrial calcium content and cytoplasmic calcium content are closely related.
