目的:检测和探讨相同遗传背景结直肠腺瘤-癌组织序列中C/EBP同源蛋白(CHOP)表达差异及其与细胞增殖/凋亡比率(PAR)之间的关系。方法:收集23例同时具有3种病理组织学类型的结直肠肿瘤(伴低级别上皮内瘤变腺瘤、伴高级别上皮内瘤变腺瘤和腺癌)病人的手术切除标本及对应正常肠黏膜,采用免疫组织化学SABC法检测CHOP表达水平及细胞增殖指数(PI,即Ki-67阳性细胞率);采用TUNEL法检测该组织标本中的细胞凋亡指数(AI)。结果:(1)相同遗传背景条件下,CHOP在结直肠腺癌组织中的阳性细胞率显著高于伴高级别上皮内瘤变腺瘤、伴低级别上皮内瘤变腺瘤和正常肠黏膜(P<0.01);伴高级别上皮内瘤变腺瘤中的阳性细胞率显著高于伴低级别上皮内瘤变腺瘤和正常肠黏膜(P<0.01),伴低级别上皮内瘤变腺瘤中的阳性细胞率显著高于正常肠黏膜(P<0.01);(2)相同遗传背景条件下,PAR在结直肠腺癌组织显著高于伴高级别上皮内瘤变腺瘤(P<0.01)、伴低级别上皮内瘤变腺瘤和正常肠黏膜(P<0.01);伴高级别上皮内瘤变腺瘤显著高于伴低级别上皮内瘤变腺瘤和正常肠黏膜(P<0.01),伴低级别上皮内瘤变腺瘤显著高于正常肠黏膜(P<0.01);(3)相同遗传背景条件下,CHOP蛋白表达水平和PAR在伴低级别上皮内瘤变腺瘤、伴高级别上皮内瘤变腺瘤和腺癌组织中呈正相关关系(P<0.01)。结论:本实验研究了相同遗传背景下结直肠腺瘤-癌组织序列中CHOP表达水平、PAR以及两者之间的关系;随着结直肠腺瘤癌变进展,CHOP表达水平及PAR逐渐升高,且两者呈正相关关系。CHOP和细胞增殖及凋亡异常可能共同参与结直肠腺瘤癌变过程。
Objective: To detect and explore the relationship between C / EBP homologous protein (CHOP) expression and cell proliferation / apoptosis ratio (PAR) in the same genetic background of colorectal adenoma-cancer tissue. METHODS: Twenty-three surgical specimens of colorectal neoplasms with low-grade intraepithelial neoplasia accompanied by high-grade intraepithelial neoplasia adenocarcinoma and three types of histopathology were collected, Mucosa. The expression of CHOP and the proliferation index (PI) of Ki-67 positive cells were detected by immunohistochemical SABC method. The apoptosis index (AI) of Tissue samples was detected by TUNEL method. Results: (1) Under the same genetic background, the positive rate of CHOP in colorectal adenocarcinoma was significantly higher than that in patients with high grade intraepithelial neoplasia, low grade intraepithelial neoplasia and normal intestinal mucosa P <0.01). The positive cell rate in high grade intraepithelial neoplasia adenocarcinoma was significantly higher than that in low grade intraepithelial neoplasia adenoma and normal intestinal mucosa (P <0.01), with low grade intraepithelial neoplasia adenoma (P <0.01). (2) Under the same genetic background, the expression of PAR in colorectal adenocarcinoma was significantly higher than that in patients with high grade intraepithelial neoplasia (P <0.01) , With low grade intraepithelial neoplasia adenoma and normal intestinal mucosa (P <0.01). The high grade intraepithelial neoplasia adenocarcinoma was significantly higher than those with low grade intraepithelial neoplasia adenoma and normal intestinal mucosa (P <0.01) , With low-grade intraepithelial neoplasia adenocarcinoma was significantly higher than that of normal intestinal mucosa (P <0.01); (3) Under the same genetic background, CHOP protein expression and PAR in low grade intraepithelial neoplasia with high Grade intraepithelial neoplasia adenocarcinoma and adenocarcinoma showed a positive correlation (P <0.01). CONCLUSIONS: This study investigated the relationship between CHOP expression and PAR in colorectal adenoma-cancer tissue with the same genetic background. The expression of CHOP and PAR gradually increased with the progression of colorectal adenoma, And there is a positive correlation between the two. CHOP and abnormal cell proliferation and apoptosis may be involved in colorectal adenoma carcinogenesis.
