H-Y antigen is a secondary histocompatibility antigen found in mammals, birds, amphibians, teleosts, and invertebrate sex-matched cell-surface providers. It is considered to be a mammalian determinant of males because it appears only in males of mammals. To determine the molecular determinants of H-Y seropositivity, H-Y positive cells are first treated differently to alter intramolecular protein or sugar structure and then tested for ability to absorb H-Y antisera. This article aims to demonstrate that the serological determinant of H-Y antigen is sugar. To test the structural dependence of the H-Y serological activity on the polypeptide, the absorbing cells were first treated with trypsin, a-chymotrypsin and thermolysin.
