目的研究人胃癌细胞株和胃癌组织中胃肠富集Kruppel样因子(GKLF)、肠道富集Kruppel样因子(IKLF)的表达及其临床意义。方法采用免疫细胞化学法检测人正常胃上皮细胞株GES-1及胃癌细胞株MKN28、SGC7901、BGC823、HGC27中GKLF、IKLF的表达;采用免疫组织化学法检测96例胃癌组织及其远端正常胃组织中GKLF、IKLF的表达。结果GKLF在人正常胃上皮细胞株内阳性表达,在胃癌细胞株内无表达;IKLF在人正常胃上皮细胞株内无表达,在胃癌细胞株内阳性表达。胃癌组织中GKLF阳性率为20.8%,显著低于远端正常胃组织(64.6%,P<0.01),胃癌组织中IKLF阳性率为71.9%,显著高于远端正常胃组织(21.9%,P<0.01)。GKLF、IKLF表达与胃癌患者的年龄、性别、分化程度、Lauren分型无关(P>0.05),但与淋巴结是否有转移及TNM分期显著相关(P<0.01)。结论GKLF、IKLF在胃癌发生、发展中起一定的作用,检测GKLF、IKLF的表达有助于判断胃癌患者的病情程度和预后。
Objective To study the expression of Grufl-like factor (GKLF) and Kruppel-like factor (IKLF) in gastric cancer cell lines and gastric cancer tissues and their clinical significance. Methods Immunocytochemistry was used to detect the expression of GKLF and IKLF in human normal gastric epithelial cell line GES-1 and gastric cancer cell lines MKN28, SGC7901, BGC823 and HGC27. Immunohistochemistry was used to detect the expression of GKLF and IKLF in 96 gastric cancer tissues and their distal normal stomach GKLF, IKLF expression in tissues. Results GKLF was positively expressed in normal human gastric epithelial cell line but not in gastric cancer cell line. IKLF was not expressed in human normal gastric epithelial cell line and was positive in gastric cancer cell line. The positive rate of GKLF in gastric cancer was 20.8%, which was significantly lower than that in distal normal gastric tissue (64.6%, P <0.01). The positive rate of IKLF in gastric cancer was 71.9%, significantly higher than that in distal normal gastric tissue (21.9%, P <0.01). The expression of GKLF and IKLF was not related to the age, sex, differentiation, Lauren classification of patients with gastric cancer (P> 0.05), but significantly correlated with lymph node metastasis and TNM stage (P <0.01). Conclusions GKLF and IKLF play roles in the development and progression of gastric cancer. To detect the expression of GKLF and IKLF is helpful to judge the severity and prognosis of patients with gastric cancer.
