目的探讨雷帕霉素对耐药胃癌细胞对多柔比星敏感性的影响。方法用含低浓度(0.5μmol/L)多柔比星培养基培养并建立耐药胃癌细胞株MKN45-R和SNU1-R,以不同浓度的多柔比星处理胃癌细胞48h后,用CCK-8试剂盒测得细胞活力,计算半数抑制浓度(IC_(50))。采用10μmol/L雷帕霉素预处理耐多柔比星胃癌细胞后再次测定IC_(50)。结果多柔比星对胃癌细胞株MKN45和SNU1的生长有剂量依赖性抑制作用,IC_(50)分别为(57.2±4.0)μmol/L和(49.9±4.7)μmol/L,耐药细胞株MKN45-R和SNU1-R的多柔比星IC_(50)显著增大,分别为(153.7±6.7)μmol/L和(172.8±13.1)μmol/L。低浓度雷帕霉素可增加耐药细胞MKN45-R和SNU1-R对多柔比星的敏感性,IC_(50)显著减小为(69.9±2.3)μmol/L和(50.0±1.7)μmol/L。结论雷帕霉素可增加耐药胃癌细胞对多柔比星的敏感性。
Objective To investigate the effect of rapamycin on drug sensitivity of gastric cancer cells to doxorubicin. Methods The gastric cancer cell lines MKN45-R and SNU1-R were cultured and cultured in low concentration (0.5μmol / L) doxorubicin medium. The gastric cancer cells were treated with different concentrations of doxorubicine for 48h, 8 kit measured cell viability, calculate the median inhibitory concentration (IC_ (50)). IC50 (50) was determined again after pretreatment with 10 μmol / L rapamycin for the anti-Doxorubicin gastric cancer cells. Results Doxorubicin inhibited the growth of gastric cancer cell lines MKN45 and SNU1 in a dose-dependent manner. The IC 50 values were (57.2 ± 4.0) μmol / L and (49.9 ± 4.7) μmol / L, respectively. The values of doxorubicin IC 50 increased significantly (153.7 ± 6.7) μmol / L and (172.8 ± 13.1) μmol / L, respectively. Low concentration of rapamycin could increase the sensitivity of drug-resistant cells MKN45-R and SNU1-R to doxorubicin, IC 50 significantly decreased to (69.9 ± 2.3) μmol / L and (50.0 ± 1.7) μmol / L. Conclusion Rapamycin can increase the sensitivity of drug-resistant gastric cancer cells to doxorubicin.
