目的:研究匙羹藤Gymnema sylvestre(Retz.)Schult叶中化学成分。方法:应用硅胶、ODS柱色谱和制备HPLC等方法进行分离纯化,应用化学方法结合1H-NMR和13C-NMR波谱技术鉴定化合物结构,测定化合物的体外蛋白非酶糖基化抑制活性,其中化合物4在50 mg·L-1时有较好抑制作用,抑制率为42.11%。结果:从匙羹藤叶中分离得到5个化合物,分别为灰毡毛忍冬皂苷乙(1)、灰毡毛忍冬皂苷甲(2)、lonimacranthoide III(3)、朝霍定C(4)、3-O-β-D-吡喃葡萄糖基-(1→3)-α-L-吡喃鼠李糖基-(1→2)-α-L-吡喃阿拉伯糖基-常春藤皂苷元-28-O-β-D-吡喃葡萄糖基酯(5)。评价了化合物1~5的体外蛋白非酶糖基化抑制活性。结论:化合物1~5均为首次从本属植物中分离得到,体外活性筛选表明化合物4在高浓度时具有一定的蛋白非酶糖基化抑制作用。
Objective: To study the chemical composition of gymnema sylvestre (Retz.) Schult leaf. Methods: The compounds were isolated and purified by silica gel, ODS column chromatography and preparative HPLC. The chemical structures of compounds were identified by 1H-NMR and 13C-NMR spectroscopy. The non-enzymatic glycosylation inhibitory activities of the compounds were determined. Compounds 4 In the 50 mg · L-1 has a good inhibitory effect, the inhibition rate was 42.11%. RESULTS: Five compounds were isolated from the leaves of Gymnema sibirica, and they were respectively identified as Lonicera saponin B (1), Lonicera saponin A (2), lonimacranthoide III (3) 3-O-β-D-glucopyranosyl- (1 → 3) -α-L-rhamnopyranosyl- (1 → 2) -α-L-arabinopyranosyl- -28-O-β-D-glucopyranosyl ester (5). In vitro protease non-enzymatic glycosylation inhibitory activity of Compounds 1-5 was evaluated. CONCLUSION: Compounds 1 to 5 are isolated from this genus for the first time. In vitro activity screening showed that compound 4 has certain non-enzymatic glycosylation inhibition at high concentration.
