目的获得能够结合细胞表面前列腺干细胞抗原(prostate stem cell antigen,PSCA)的抗人PSCA单克隆抗体。方法以重组人PSCA蛋白为免疫原,通过杂交瘤技术制备抗人PSCA单克隆抗体,并进行ELISA、Western印迹、流式细胞术等实验筛选。结果获得6株抗人PSCA单克隆抗体,经过筛选最终得到3株能够结合细胞表面PSCA的单克隆抗体。结论获得具有结合细胞表面PSCA活性的单克隆抗体,为进一步研究和应用奠定了基础。
Objective To obtain anti-human PSCA monoclonal antibody that can bind to cell surface prostate stem cell antigen (PSCA). Methods Recombinant human PSCA protein was used as an immunogen to prepare anti-human PSCA monoclonal antibody by hybridoma technique. The anti-human PSCA monoclonal antibody was tested by ELISA, Western blot and flow cytometry. Results Six anti-human PSCA monoclonal antibodies were obtained. After screening, three monoclonal antibodies capable of binding PSCA on the cell surface were obtained. Conclusion Obtaining of monoclonal antibodies with PSCA activity on the cell surface lays the foundation for further research and application.
