目的应用近红外光谱法对红花中羟基红花黄色素A进行快速测定。方法利用HPLC法测定样品中羟基红花黄色素A的量,运用偏最小二乘(PLS)法建立其量与NIR光谱之间的多元校正模型,对未知样品中的量进行预测。结果模型以0~1间归一化(n01)结合一阶导数,以偏最小二乘法(PLS)建模效果最好,校正集和预测集的相关系数分别为0.967 7和0.956 4,所建立模型具有相对较好的内部稳健性、拟合度和预测能力。结论本方法可用于红花中羟基红花黄色素的快速测定。
Objective To determine the rapid determination of hydroxysafflor yellow A in safflower by near infrared spectroscopy. Methods The amount of hydroxysafflor yellow A in the sample was determined by HPLC, and the multivariate calibration model was established by partial least squares (PLS) method to predict the amount of hydroxysafflor yellow in the sample. Results The model is best modeled by partial least squares (PLS) with first-order derivatives normalized from 0 to 1 (n01), and the correlation coefficients between the calibration set and the prediction set are 0.967 7 and 0.956 4 respectively The model has relatively good internal robustness, fitness and predictive ability. Conclusion This method can be used for the rapid determination of hydroxysafflor yellow in safflower.
