目的 探讨卡托普利及SNP对培养的大鼠VSMC释放ET 1的影响及相互关系。方法 在大鼠VSMC培养液中置入不同浓度卡托普利及 /或SNP后 ,用放免法测定ET 1。结果 卡托普利及SNP均可减低因AngⅡ引起VSMC分泌增加的内皮素量 ,AngⅡ水平与VSMC分泌ET 1量呈正相关。结论 ET 1通过抑制VSMC内源性NO的产生加速AS的发生及发展 ,ACEI或NO抑制ET 1释放 ,可减缓AS形成。
Objective To investigate the effects of captopril and SNP on ET 1 release from cultured rat VSMCs and their correlation. Methods After different concentrations of captopril and / or SNP were placed in rat VSMC medium, ET 1 was determined by radioimmunoassay. Results Both captopril and SNP could reduce the amount of endothelin induced by AngⅡ, and the level of AngⅡ was positively correlated with the amount of ET 1 secreted by VSMC. Conclusions ET 1 accelerates the occurrence and development of AS by inhibiting the production of endogenous NO in VSMC. ACEI or NO inhibits the release of ET 1 and attenuates AS formation.
