从流产的人胎皮提纯的角蛋白作为抗原,免疫BALB/C小鼠,通过杂交瘤技术获得一株稳定分泌抗角蛋白单抗(命名为ⅢG1)的杂交瘤。其染色体数为90~98条。用ABC-ELISA法检测ⅢG1单抗亚类为IgG1。用人胚胎及正常成人多组织的免疫组化法显示皮肤基底细胞、肾近曲小管和肠上皮呈阳性染色。同法检测部分肿瘤组织也呈阳性反应。表明ⅢG1针对的抗原决定簇相对较窄,可能是一株仅对低分子量角蛋白反应的单抗。对来源于上皮细胞的癌有诊断价值。
Purified keratin derived from abortive human fetal skin was used as an antigen to immunize BALB/C mice, and a hybridoma that stably secreted an anti-keratin monoclonal antibody (named IIIG1) was obtained by the hybridoma technique. Its chromosome number is 90-98. The subclass of IIIG1 monoclonal antibody was IgG1 detected by ABC-ELISA. Immunohistochemistry with multiple tissues from human embryos and normal adults showed positive staining of basal cells, renal proximal tubules and intestinal epithelium. The same method used to detect part of the tumor tissue also showed a positive reaction. It was shown that the antigenic determinants targeted by IIIG1 are relatively narrow and may be a monoclonal antibody that reacts only to low molecular weight keratin. It is of diagnostic value for cancers derived from epithelial cells.
