目的分析miR-373*、miR-200c在乳腺癌干细胞(breast cancer stem cells,BCSCs)和MCF-7乳腺癌细胞株中的差异表达。方法 Trizol法分别抽提乳腺癌干细胞和MCF-7细胞总RNA,实时定量PCR检测miRNAs表达,对表达差异显著的miRNAs进行潜在靶基因预测。结果 miR-373*在乳腺癌干细胞中较MCF-7细胞高表达(6.684±0.548),而miR-200c在乳腺癌干细胞中较MCF-7细胞低表达(0.345±0.031)。通过软件分析预测,miR-373*与TP53INP2、CASP8、EIF4A1、EEF1A1等基因均具有可能靶位点,miR-200c与TCF2、TDE2、SYVN1、PDCD10、RAP2C等基因均具有可能靶位点。结论 miR-373*在乳腺癌干细胞中高表达,可能是一个潜在的癌基因。miR-200c在乳腺癌干细胞中低表达,可能是一个潜在的抑癌基因。
Objective To analyze the differential expression of miR-373 * and miR-200c in breast cancer stem cells (BCSCs) and MCF-7 breast cancer cell lines. Methods Total RNA was extracted from breast cancer stem cells and MCF-7 cells by Trizol method. The expression of miRNAs was detected by real-time quantitative PCR, and the potential target genes were predicted for miRNAs with significant difference. Results miR-373 * was highly expressed in breast cancer stem cells compared with MCF-7 cells (6.684 ± 0.548), while miR-200c was lower than that in MCF-7 cells (0.345 ± 0.031) in breast cancer stem cells. By software analysis and prediction, miR-373 * and TP53INP2, CASP8, EIF4A1, EEF1A1 and other genes have potential target sites, miR-200c and TCF2, TDE2, SYVN1, PDCD10, RAP2C and other genes have potential target sites. Conclusion miR-373 * is highly expressed in breast cancer stem cells and may be a potential oncogene. The low expression of miR-200c in breast cancer stem cells may be a potential tumor suppressor gene.
