研究建立以反相高效液相色谱法测定环孢霉素A全血浓度的方法。方法建立简便易行的血样提取方法,采用C18柱,乙晴和水为流动相,分离药物,紫外检测器测定浓度。结果线性范围为75~1200μg/L,提取回收率和方法回收率分别为(70.4±1.5)%和(99.7±0.6)%,日内和日间变异系数分别为(1.3±0.4)%和(4.1±0.3)%,无其它常用药物的干扰。结论本方法操作简便,可用于临床血药浓度的监测。
To establish a method for the determination of cyclosporin A whole blood concentration by reversed-phase high performance liquid chromatography. Methods A simple and convenient method of blood sampling was established. C18 column, acetonitrile and water were used as the mobile phase to separate the drug and UV detector to determine the concentration. The results showed that the linear range was 75 ~ 1200μg / L, the recovery rate and recovery rate were (70.4 ± 1.5)% and (99.7 ± 0.6)%, respectively. The intra- and inter- 1.3 ± 0.4)% and (4.1 ± 0.3)%, no other commonly used drugs. Conclusion The method is simple and can be used for the monitoring of clinical blood concentration.
