目的 研究超抗原金黄色葡萄球菌肠毒素A(SEA)刺激后的树突状细胞 (DC)体内诱导的抗膀胱肿瘤免疫效应。方法 分离小鼠T739脾脏DC ,加SEA贴壁培养并富集细胞 ,观察其对DC功能特性的影响及体内诱导的抗肿瘤免疫保护反应。结果 经SEA刺激后的DC共刺激分子B7表达显著增加 ,刺激T细胞增殖能力明显增强 (P <0 .0 5 ) ;在体内诱导出更强的细胞毒性T细胞 (CTL) ,有效抵抗膀胱肿瘤细胞BTT739的攻击。结论 SEA可活化DC ,使后者在体内诱导出显著的抗膀胱肿瘤保护性免疫反应
Objective To investigate the anti-bladder tumor immune response induced by dendritic cells (DCs) stimulated by the superantigen Staphylococcus aureus enterotoxin A (SEA). Methods T739 splenic DCs were isolated from mice and cultured with SEA. The effects of DCs on the functional properties of DCs and the anti-tumor immune protective responses in vivo were observed. Results The expression of B7, a costimulatory molecule stimulated by SEA, was significantly increased and the ability of stimulating T cell proliferation was significantly enhanced (P <0.05). More powerful cytotoxic T lymphocytes (CTL) were induced in vivo, which effectively prevented bladder cancer Cell BTT739 attack. Conclusions SEA can activate DC, which induces a significant protective immunity against bladder tumor in vivo
