目的探讨碱性成纤维生长因子(bFGF)、血管内皮生长因子(VEGF)在低氧性肺动脉高压(HPH)大鼠肺组织中表达的意义。方法雄性SD大鼠24只,随机分成对照组和实验组,干预后,右心导管法测各组大鼠肺血流动力学指标,称重法测右室肥厚指标(RVHI);光镜观察肺血管病理改变;逆转录聚合酶链反应(RT-PCR)检测bFGFmRNA和VEGFmRNA。结果对照组大鼠的平均肺动脉压(mPAP)和RVHI值显著低于实验组大鼠(P<0.01);光镜显示,实验组大鼠肺血管壁病理学变化较对照组显著。RT-PCR结果提示,实验组大鼠肺组织中bFGF和VEGF值与对照组比较显著升高(P<0.05)。结论低氧可引起bFGF和VEGF的合成增多,bFGF和VEGF参与了低氧性肺动脉高压的发病过程。
Objective To investigate the expression of basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF) in lung tissue of rats with hypoxic pulmonary hypertension (HPH). Methods Twenty-four male Sprague-Dawley rats were randomly divided into control group and experimental group. After intervention, the indexes of pulmonary hemodynamics of rats in each group were measured by right heart catheterization, and the right ventricular hypertrophy index (RVHI) Pulmonary vascular pathological changes; reverse transcription polymerase chain reaction (RT-PCR) detection of bFGFmRNA and VEGFmRNA. Results The mean pulmonary arterial pressure (mPAP) and RVHI in the control group were significantly lower than those in the experimental group (P <0.01). The light microscopy showed that the pathological changes of the pulmonary vascular wall in the experimental group were significantly more than those in the control group. The results of RT-PCR indicated that the bFGF and VEGF values in the lung tissue of the experimental group were significantly increased compared with the control group (P <0.05). Conclusion Hypoxia can induce the synthesis of bFGF and VEGF, and bFGF and VEGF are involved in the pathogenesis of hypoxic pulmonary hypertension.
